Scientists grow "mini human brains" from stem cells

LONDON (Reuters) - Scientists have grown the first mini human brains in a laboratory and say their success could lead to new levels of understanding about the way brains develop and what goes wrong in disorders like schizophrenia and autism.

Researchers based in Austria started with human stem cells and created a culture in the lab that allowed them to grow into so-called "cerebral organoids" - or mini brains - that consisted of several distinct brain regions.

It is the first time that scientists have managed to replicate the development of brain tissue in three dimensions.

Using the organoids, the scientists were then able to produce a biological model of how a rare brain condition called microcephaly develops - suggesting the same technique could in future be used to model disorders like autism or schizophrenia that affect millions of people around the world.

"This study offers the promise of a major new tool for understanding the causes of major developmental disorders of the brain ... as well as testing possible treatments," said Paul Matthews, a professor of clinical neuroscience at Imperial College London, who was not involved in the research but was impressed with its results.

Zameel Cader, a consultant neurologist at Britain's John Radcliffe Hospital in Oxford, described the work as "fascinating and exciting". He said it extended the possibility of stem cell technologies for understanding brain development and disease mechanisms - and for discovering new drugs.

Although it starts as relatively simple tissue, the human brain swiftly develops into the most complex known natural structure, and scientists are largely in the dark about how that happens.

This makes it extremely difficult for researchers to gain an understanding of what might be going wrong in - and therefore how to treat - many common disorders of the brain such as depression, schizophrenia and autism.

GROWING STEM CELLS

To create their brain tissue, Juergen Knoblich and Madeline Lancaster at Austria's Institute of Molecular Biotechnology and fellow researchers at Britain's Edinburgh University Human Genetics Unit began with human stem cells and grew them with a special combination of nutrients designed to capitalize on the cells' innate ability to organize into complex organ structures.

They grew tissue called neuroectoderm - the layer of cells in the embryo from which all components of the brain and nervous system develop.

Fragments of this tissue were then embedded in a scaffold and put into a spinning bioreactor - a system that circulates oxygen and nutrients to allow them to grow into cerebral organoids.

After a month, the fragments had organized themselves into primitive structures that could be recognized as developing brain regions such as retina, choroid plexus and cerebral cortex, the researchers explained in a telephone briefing.

At two months, the organoids reached a maximum size of around 4 millimeters (0.16 inches), they said in a report of their study published in the journal Nature.

Although they were very small and still a long way from resembling anything like the detailed structure of a fully developed human brain, they did contain firing neurons and distinct types of neural tissue.

"This is one of the cases where size doesn't really matter," Knoblich told reporters.

"Our system is not optimized for generation of an entire brain and that was not at all our goal. Our major goal was to analyze the development of human brain (tissue) and generate a model system we can use to transfer knowledge from animal models to a human setting."

In an early sign of how such mini brains may be useful for studying disease in the future, Knoblich's team were able to use their organoids to model the development of microcephaly, a rare neurological condition in which patients develop an abnormally small head, and identify what causes it.

Both the research team and other experts acknowledged, however, that the work was a very long way from growing a fully-functioning human brain in a laboratory.

"The human brain is the most complex thing in the known universe and has a frighteningly elaborate number of connections and interactions, both between its numerous subdivisions and the body in general," said Dean Burnett, lecturer in psychiatry at Cardiff University.

"Saying you can replicate the workings of the brain with some tissue in a dish in the lab is like inventing the first abacus and saying you can use it to run the latest version of Microsoft Windows - there is a connection there, but we're a long way from that sort of application yet."

Saturn Moon Titan Sports Thick Icy Shell & Bizarre Interior




" The tough icy shell of Saturn's largest moon Titan is apparently far stronger than previously thought, researchers say.

These surprising new findings add to hints Titan possesses an extraordinarily bizarre interior, scientists added.

Past research suggested Titan has an ocean hidden under its outer icy shell 30 to 120 miles (50 to 200 kilometers) thick. Investigators aim to explore this underground ocean in the hopes of finding alien life on Titan, since virtually wherever there is water on Earth, there is life. To learn more about Titan's icy shell, planetary scientist Doug Hemingway at the University of California, Santa Cruz, analyzed the Cassini probe's scans of Titan's gravity field. The strength of the gravitational pull any point on a surface exerts depends on the amount of mass underneath it. The stronger the pull, the more the mass.

The researchers then compared these gravity results with the structure of Titan's surface. They expected that regions of high elevation would have the strongest gravitational pull, since one might suppose they had extra matter underneath them. Conversely, they expected regions of low elevation would have the weakest gravitational pull.

What the investigators discovered shocked them. The regions of high elevation on Titan had the weakest gravitational pull.

"It was very surprising to see that," Hemingway told SPACE.com. "We assumed at first that we got things wrong, that we were seeing the data backwards, but after we ran out of options to make that finding go away, we came up with a model that explains these observations."

To explain these gravity anomalies, Hemingway said to imagine mountains on Titan having roots. "It's like how most of an iceberg actually lies submerged underwater," he said. "If that root is really big, bigger than normal, it would displace water underneath it."

Ice has a lower density than water — a chunk of ice weighs less than a similar volume of water. These high-elevation areas on Titan apparently have roots large enough to displace a lot of water under them, meaning they exert a weaker gravitational pull.

Ice is buoyant in water. "In order to essentially hold these big icebergs down and keep them from bobbing up, that means Titan's shell has to be extremely rigid," Hemingway said.

It remains uncertain what makes Titan's shell this rigid. The ice might possess cage-like molecules known as clathrates that could make it stiffer. Also, "if the ocean underneath the shell is colder than before thought, that could make the ice shell thicker and thus more rigid," Hemingway said.

This rigidity could mean Titan's shell is less geologically active than once thought. "If at least the top 40 kilometers (25 miles) is very stiff and cold and dead, if you want something like cryovolcanoes that erupt water instead of lava on Titan's surface, you have to be more creative about how that might happen," Hemingway said.

Their model also suggests Titan's shell has seen an extensive amount of erosion, with features carved more than 650 feet (200 meters) deep on it surface. "We now need different groups of people to figure out how so much material could get broken up and transported long distances across Titan's surface," Hemingway said.

One implication of these new findings relates to whether or not Titan's interior is separated into distinct layers. If researchers have underestimated Titan's gravity field, one might suspect its core is a giant blob of matter that is not made up of distinct layers as one would expect from such a large body. For instance, Earth is separated into a crust, mantle and core, and even large asteroids such as Vesta seem to have interiors divided into several layers.

"Maybe Titan is a mixture of ice and rock from the core nearly all the way out, and it's only in the last part near its surface that it's differentiated into ice and water," Hemingway said. "But we could be wrong there."

To help solve this mystery, "what we need is a Titan orbiter," Hemingway said. "That way we can have much better readings of Titan and learn more about its ice shell and its interior."

The scientists detailed their findings in the Aug. 29 issue of the journal Nature.

Las Vegas casino seeks to evict raunchy nightclub




"LAS VEGAS (AP) — It turns out that even in Sin City, some sins are hard to overlook.

The Palazzo hotel-casino on the Las Vegas Strip is trying to evict a 10-month-old nightclub for shows it says are so raunchy that they violate obscenity laws. It says actors — some nearly naked — toss condoms into the crowd and simulate sex acts and bestiality on stage.

The club is seeking a restraining order to halt the closure, arguing that simulated sex acts don't constitute obscenity.

Casino officials "were well aware of our brand," said Sean Dunn, special events director at The Act, in an email statement, adding that representatives of the hotel-casino have frequently attended shows and did not complain.

Las Vegas Sands, which owns the casino, did not immediately respond to requests for comment.

The Act remains open for business while a district judge considers its fate, but the fight over its future has exposed an underlying reality in Vegas: While the city sells itself as a racy, no-holds-barred destination, there are limits.

"I think there's the perception that anything goes in Vegas — there's no boundaries, no lines," said Lynn Comella, a professor of sexuality and women's studies at the University of Nevada, Las Vegas. "In reality, nothing could be further from the truth."

That perception, in part, is fueled by what tourists see.

Trucks roll down Las Vegas Boulevard advertising "hot babes" delivered directly to hotel rooms, the phone book is full of come-ons for escorts and private dancers, and taxis and billboards feature scantily clad women who leave little to the imagination.

While Las Vegas is also rife with strip clubs, tourists won't find them along the Strip or in the city's hotel-casinos. They can take in a topless stage show or lounge at a European-style topless pool, but they have to catch a ride to a side street to see strippers.

"The goal for Las Vegas was to be naughty enough to attract you but not naughty enough to repel you," said Michael Green, a history professor at the College of Southern Nevada.

The city — which rises and falls on the pocketbooks of its 40 million annual visitors — must balance the sexiness with the sensibilities of foreign tourists and large, multinational casinos.

"It's highly sexualized and gets a lot of mileage out of that, but it's a very particular version of that," Comella said.

In The Act's case, Sands executives notified club officials on April 26 that they'd crossed a line with the show and were no longer welcome in the $15 million space. The casino giant wants to evict it, ending a 10-year lease.

The casino notified club officials after undercover security officers went into the venue in March. The details of the show were laid bare this week in court, when an investigator apologized for getting graphic and looked pained while describing some kinky scenes.

The club contends Sands "manufactured this 'breach' in an attempt to improperly evict" it.

"We believe we are on the leading edge of the next evolution of nightlife in Las Vegas and that The Act fills a niche for patrons looking for an alternative to the typical Las Vegas big-box club scene," Dunn said.

Those who stray outside the limits of what is considered acceptable in the city risk invoking the wrath of the state's powerful Gaming Control Board and losing their lucrative gambling permits.

The board smacked the Planet Hollywood casino with a $750,000 fine in 2009 amid accusations that employees at the independently owned Prive nightclub allowed toplessness, turned a blind eye to prostitution and physically and sexually assaulted patrons.

The board isn't investigating The Act and is waiting for the lawsuit to play out, according to chairman A.G. Burnett.

There's no doubt that Las Vegas markets itself as an adult playground, said Oscar Goodman, the larger-than-life former Las Vegas mayor who's often flanked by scantily clad showgirls, martini in hand.

"We have standards and we're not going to advocate illegal activity," he said. "Unless the sex goes out of the line, we embrace it.""

Watch 'Duck Dynasty' Star Phil Robertson's Antiabortion Speech: 'You Have a God-Given Right to Live'




"The family members in A&E's monster hit reality series "Duck Dynasty" have always been upfront but not in-your-face about their faith on camera. Off camera, though, the Robertsons are not afraid to loudly proclaim their religious beliefs, including their stance against abortion.

Video surfaced Tuesday of "Duck Dynasty" patriarch Phil Robertson delivering an impassioned speech slamming abortion. "From the time you started inside your mother's womb, Thomas Jefferson had it right," he said. "You have the God-given right to live, for crying out loud."

He continued by holding out a finger. "You're this long! C'mon! You have a God-given right to live. And of all places, inside your mother — what in the world happened to us?"

Robertson, a former standout football quarterback at Louisiana Tech, has been an ardent believer since the '70s. During that time, he "was headed south" — drinking, fighting, mistreating his family. When his wife, Kay, kicked him out, Robertson turned to religion.

Though Robertson feels free to let loose his opinions in a speech, he told the Wall Street Journal in June that his family purposefully plays down their faith on "Duck Dynasty."

"We can't get into spiritual matters on the show too much," he said. "That's a little much for the production company. They say, you know, it's not the Pat Robertson show."

Still, the family doesn't hide their beliefs. And it's possible religion will play a greater role this season, with oldest son Alan — a former minister — joining the cast. In fact, before the season began, Alan said he hoped to spread his message to a bigger audience thanks to the popularity of "Duck Dynasty."

Alan said of leaving his pulpit, "What I do for our church — you see, it's a pretty good-sized church — impacts a lot of people. But because of my association with the show, I'll get to minister to a lot more people.""





367 Pit Bulls Rescued in Multi-State Dog Fighting Bust




" A three-year investigation and multi-state dog fighting bust resulted in the rescue of 367 pit bull terriers in Alabama and Georgia, according to officials.

"The dogs, ranging in age from just several days to 10-12 years, had been left to suffer in extreme heat with no visible fresh water or food," the ASPCA said in a statement. "Many are emaciated with scars and wounds consistent with dog fighting, and some were tethered by chains and cables that were attached to cinder blocks and car tires."

The bust also included the arrest and indictment of 10 people from Alabama, Texas, Georgia and Mississippi for violating the federal dog fighting statute and the federal gambling statute, according to the U.S. Attorney's Office in Alabama.

Authorities also seized guns, illegal narcotics, drugs used to treat and train dogs and $500,000 from the dog fighters involved in the organization.

The defendants were betting between $5,000 and $200,000 on each fight, according to U.S. Attorney George Beck, Jr.

"The number of dogs seized and the amount of money involved in this case shows how extensive this underworld of dog fighting is," Beck said in a statement. "These dog fighters abuse, starve and kill their dogs for the supposed 'fun' of watching and gambling on a dog fight. Their behavior is deplorable, will not be tolerated, and will be punished to the full extent of the law."

In addition to the ASPCA and the U.S. Attorney's Office, the operation included the Auburn Police, FBI and the Humane Society of the United States.

The dogs were taken to temporary emergency shelters in undisclosed locations where they are receiving treatment, according to the ASPCA.

"Today we ended the torture of hundreds of abused and neglected dogs," ASPCA president and CEO Matt Bershadker said in a statement. "Never again will these dogs be forced to fight, live in squalor, or be neglected and deprived of the bare necessities."

When asked about the possibility of adopting out the dogs, ASPCA spokeswoman Alison Jimenez told ABCNews.com in an email, "It's too early to determine placement for these dogs since evaluations have yet to be conducted. Additionally, this is an ongoing investigation and the animals are considered evidence until legally turned over to the ASPCA.""

Lamar Surfaces, Khloé Puts on Ring … but Will That Hush Those Divorce Rumors?




"Call off the missing-persons-alert: Lamar Odom has returned home and is talking again with wife Khloé Kardashian.

Amid rampant reports that his five-year marriage is on the rocks, Lamar surfaced on Monday afternoon in the San Fernando Valley. It's the first time the NBA star has been seen since publicly last Wednesday.

Odom was photographed in a white SUV near the home he shares with Khloé, and was later seen driving into their gated community.

In a positive sign that the "Keeping Up With the Kardashians" twosome is trying to work things out, Lamar was photographed with his wedding ring on. X17online also spotted Khloé, who had ditched her band last week on a trip to the gym, sporting her ring on Monday afternoon post-workout.

The Kardashian family has been concerned about Lamar. Several sources close to the clan have confirmed to omg! that the basketballer, who played with the Los Angeles Clippers last season, was struggling with drug abuse. However, the insiders insisted that the family knew where Lamar was and that he was not missing as some reports had claimed.

Odom, 33, has acknowledged smoking pot in the past. Sources tell omg! that he has recently moved on to harder drugs.

TMZ, which first broke news of Odom's latest personal struggles, reported on Tuesday that Odom's worsening drug use affected his 2012 basketball season with the Dallas Mavericks. All indications are that Odom was clean during his past season with the Clippers but then fell back on bad habits once the season ended in June. Tabloid cover stories have also claimed that Lamar was cheating on Khloé, which they have both denied.

However, according to Us Weekly, Khloé, 29, had been hiding her marital problems from her family before things exploded last week.

Still, the couple is trying to put down reports of imminent divorce.

In a Tuesday story, E! News quoted a family source as saying, "They have never spoken to a divorce attorney.

"Right now the family is focused on getting Lamar better.""





Alec Baldwin Involved in Paparazzi Altercation




"Alec Baldwin was involved in an altercation with a paparazzi in New York City today, his latest confrontation with the ubiquitous photo-takers who track the moves of celebrities.

According to TMZ, which first reported the news, the photographer, who has not been named in the account, nor by authorities, claimed that he did not initiate the fight. "All I can say is I was doing my job asking him normal questions and he flipped," he said. "That's all."

However, a source said the opposite was true.

"The photographer was the aggressor," the source told ABC News. "He even chased Alec's wife [Hilaria] into a store after the incident."

The website later reported that both parties admitted to pushing each other.

When contacted by ABC News, Lt. Karen Anderson of the NYPD said that "nobody got hurt" and that no police report was filed.

A rep for Baldwin had no comment.

Baldwin, 55, and his wife, 29, welcomed a daughter, Carmen Gabriela, in New York City last Friday.

'Very little doubt' chemical weapons used in Syria




"The United States said Sunday there was "very little doubt" Syrian forces had used chemical weapons on civilians and dismissed an offer by Damascus for a UN team to view the attack site.

The comments marked a significant escalation of a showdown over the horrific attack outside the Syrian capital that killed up to 1,300 people last week, and came as Washington appeared to be positioning for possible military action.

Officials said President Barack Obama, who held crisis talks Saturday with top aides, would make an "informed decision" about how to respond to an "indiscriminate" chemical weapons attack.

An official told AFP that, based on the reported number of victims and their symptoms and on US and foreign intelligence, "there is very little doubt at this point that a chemical weapon was used by the Syrian regime against civilians in this incident."

The official, speaking on condition of anonymity, said Washington had noted that Syria had offered to let UN inspectors view the site of the alleged attack on Monday, but suggested it was too little, too late.

"If the Syrian government had nothing to hide and wanted to prove to the world that it had not used chemical weapons in this incident, it would have ceased its attacks on the area and granted immediate access to the UN five days ago," the official said.

"At this juncture, the belated decision by the regime to grant access to the UN team is too late to be credible, including because the evidence available has been significantly corrupted as a result of the regime's persistent shelling and other intentional actions over the last five days."

The United Nations meanwhile said that its team would start a probe on Monday into what happened in the area, which is under rebel control.

President Bashar al-Assad's government said the visit would prove claims by the opposition that chemical weapons were used against defenseless civilians, including children, were "lies."

The opposition says 1,300 people were killed. Doctors Without Borders says 355 people died in hospital alone from "neurotoxic" symptoms.

Several factors -- including a US diplomatic offensive led by Secretary of State John Kerry, comments from the White House and signs the Pentagon is positioning ships closer to Syria -- fueled an impression Obama may be preparing to jettison his antipathy to new Middle Eastern entanglements and order limited military action.

But a White House official shot down a report in Britain's Telegraph newspaper claiming that London and Washington planned to join forces and launch military action against Syria "within days."

"The president has not made a decision to undertake military action," the official said.

French President Francois Hollande told Obama on Sunday in a phone call that "everything was consistent" with the conclusion that Damascus was behind last Wednesday's suspected chemical attack.

"The two presidents agreed to stay in close contact to arrive at a joint response to this unprecedented aggression," the French government said in a statement.

Russia, however, reacted to US maneuvering by warning Washington against making a "tragic mistake."

Polls show Americans are wary of getting into another war in the Middle East.

But Obama's credibility is on the line after he said last year that the use of large-scale chemical weapons in Syria would cross a US "red line."

Washington may also want to send a signal that weapons of mass destruction, like chemical arms, cannot be used in Syria, or elsewhere, with impunity.

Republican senators John McCain and Lindsey Graham, who have criticized Obama's failure to use military force to protect civilians in a war that has killed 100,000 people, upped pressure on the president on Sunday.

"The United States must rally our friends and allies to take limited military actions in Syria that can change the balance of power on the ground and create conditions for a negotiated end to the conflict and an end to Assad's rule," the senators said in a statement.

"Using stand-off weapons, without boots on the ground, and at minimal risk to our men and women in uniform, we can significantly degrade Assad's air power and ballistic missile capabilities and help to establish and defend safe areas on the ground."

Experts say the most likely US action would involve sea-launched cruise missiles targeting Syrian military installations and artillery batteries deemed complicit in the chemical weapons attack.

Weapons fired from US planes outside the country could also be used, to minimize the risk to US or allied pilots from Syrian air defenses.

Washington would likely seek to act with a broad coalition of European and Gulf allies as Russia is seen sure to veto any attempt in the UN Security Council to mandate action against its Middle Eastern ally.

Obama called British Prime Minister David Cameron on Saturday to discuss options, after a meeting of all his top diplomatic, military and intelligence chiefs at the White House.

Kerry has spent days on the phone with Washington's foreign partners.

US Defense Secretary Chuck Hagel said on a tour of Asia the military would be ready to act if called on by Obama.

"President Obama has asked the Defense Department to prepare options for all contingencies. We have done that," Hagel said."

"Hot duo alert! Miley Cyrus is set to collaborate with Robin Thicke to his smash hit "Blurred Lines" at the MTV Video Music Awards Sunday, sources confirm exclusively to Us Weekly.

"They're friends and she loves the song," a source tells Us. A second insider adds, "It's amazing. She comes out of a giant bear wearing a very provocative outfit." The prop is perhaps paying tribute to her raunchy "We Can't Stop" music video in which she danced suggestively with massive stuffed bears.

"Today's already the f-cking best day ever! Just wait till VMAs tonight," the 20-year-old star wrote via Twitter hours before the big show. For his part, 36-year-old Thicke later tweeted, "Who's ready for my performance at the @MTV #VMA's tonight? #VoteThicke."

Cyrus is nominated for Best Female Video, Video of the Year and Best Editing for "We Can't Stop." Thicke's "Blurred Lines" featuring T.I. and Pharrell is in the running for Video of the Year, Best Male Video and Best Collaboration.

Jenna Wolfe and Stephanie Gosk Are First-Time Mommies!




"Breaking news! Or at least it is for "Today" anchor Jenna Wolfe and her partner, NBC News foreign correspondent, Stephanie Gosk, who welcomed a pink little bundle of joy into their family on Wednesday.

The baby girl, Harper Estelle Wolfeld Gosk, arrived at 7:31 p.m. weighing 7 pounds, 13 ounces. This is the first child for both mommies.

"We couldn't be happier. Thanks for all the great messages," Gosk, 40, tweeted.

"Fell madly in love w Harper Estelle tonight," Wolfe, 39, added on the social network. "Thx for the kind words and well wishes. Other than that I'm speechless."

The couple, who is not married, announced that they were expecting on March 27 — the same day that they came out as a couple.

"My girlfriend, Stephanie Gosk, who works here at NBC, and I are expecting a baby girl at the end of August," Wolfe told Matt Lauer on the "Today" show in March. "Stephanie's extremely adventurous, she's been all over the world. And you know me -- there's not a building I haven't jumped off of … so, for us, this is nothing like that.
